Analogous to ubiquitin, reversible covalent attachment of SUMO (small ubiquitin-like modifier) to lysine residues in substrate proteins alters the properties of the proteins to which SUMO conjugates. The proteomic implications of protein sumoylation are reviewed in this article. A: Epitope exposure can be pH dependent, with the large majority of epitopes showing increased retrieval with increasing pH. An increase of pH from 6 to 9 can give a four-fold enhancement in staining for some epitopes. Tris/EDTA is a good high-pH antigen retrieval buffer for IHC.
