The Ubiquitin Proteasome Pathway: a ubiquitous drug target?

Review Article - A Nobel prize and an FDA-approved drug later, the ubiquitin proteasome pathway is one of the hottest fields at the moment. As the principal mechanism for protein catabolism in the mammalian cytosol and nucleus, the pathway affects a wide variety of cellular processes through E1 activating enzymes, E2/E3 ligases, and deubiquitination enzymes. This review will highlight some of the latest advances in this field and its potential impact on clinicians.

Tumor Necrosis Factor Promotes Runx2 Degradation through Up-regulation of Smurf1 and Smurf2 in Osteoblasts ™ Dr. Lianping Xing and his collaborators from the University of Rochester and NCI in Bethesda described that osteoblast differentiation is inhibited by TNF via the ubiquitin proteasome pathway. The upregulated Smurf1, an E3 ubiquitin ligase, increased degradation of the Runx2, a critical transcription factor for bone-forming osteoblast activities. The study used Abgent™s antibody to Smurf1 (Cat #AP2104a)

EpiShot™: a Rational Approach for Epitope Prediction

Featured Article - Antibodies are powerful and universal tools in today™s biology research. Outcomes of multiple-year projects often depend on the quality of the antibodies. Although producing an antibody is as easy as subcloning a gene, there are a number of conditions that go into designing the antigen and producing the antibody. Understanding those issues while working with the antibody production team enhances the likelihood of success. This multi-field expertise has been offered to our customers through our EpiShot™ epitope prediction procedure, and is now the objective of this review. Read Full Article

The Role of Branchial Carbonic Anhydrase in Acid-base Regulation in Rainbow Trout ™ A project led by Dr. S.F. Perry from the University of Ottawa (Canada) described the key catalytic activity and cellular localization of the cytoplasmic carbonic anhydrase (tCAc) in regulating the acid-base balance in rainbow trout. The custom-made antibody produced at Abgent has generated valuable data. Review Data

Custom Antibody Production ™ Abgent™s expertise in polyclonal and monoclonal antibody production is available for your on-going scientific research. A talk with our team prior starting your antibody production can save you from recurrent mistakes in antigen design. Our flier helps you to choose the project that best fits your needs. View Flier

Q: How do I know whether the bands I detect on my Western blot are specific?

A: The antigen used to raise the antibody can be used as an inhibitor to compete for antibody binding with the endogenous target. Although more validation may be required, adding a blocking peptide to the antibody preparation can solve questions about a noisy Western blot. Abgent offers Blocking Peptides complementary to Antibody products. View our Blocking Peptide protocol.
