The Ubiquitin Proteasome Pathway: a ubiquitous drug target? |
Review Article
- A Nobel prize and an FDA-approved drug later, the ubiquitin proteasome pathway is one of the hottest fields at the moment. As the principal
mechanism for protein catabolism in the mammalian cytosol and nucleus, the pathway affects a wide variety of cellular processes through E1
activating enzymes, E2/E3 ligases, and deubiquitination enzymes.
This review will highlight some of the latest advances in this field and its potential impact on clinicians.

Tumor Necrosis Factor Promotes Runx2 Degradation through Up-regulation of Smurf1 and Smurf2 in Osteoblasts ™ Dr. Lianping Xing and his
collaborators from the University of Rochester and NCI in Bethesda described that osteoblast differentiation is inhibited by TNF via the
ubiquitin proteasome pathway. The upregulated Smurf1, an E3 ubiquitin ligase, increased degradation of the Runx2, a critical transcription
factor for bone-forming osteoblast activities. The study used Abgent™s antibody to Smurf1

Q:
How do I know whether the bands I detect on my Western blot are specific?
A: The antigen used to raise the antibody can be used as
an inhibitor to compete for antibody binding with the endogenous target. Although more validation may be required, adding a blocking
peptide to the antibody preparation can solve questions about a noisy Western blot. Abgent offers Blocking Peptides complementary
