Review Article - Histone deacetylases (HDACs) are a family of enzymes whose primary function is to remove the acetyl group from histones, which causes the surrounding DNA to wind more tightly around the histone, thus denying the cell’s transcriptional machinery access to genetic material. Because HDACs play a critical role in gene expression, its dysfunction and inappropriate recruitment have been implicated in numerous diseases, ranging from leukemia to AIDS. Thus, new and previously approved drugs that inhibit HDACs have become the subject of ongoing clinical trials. Read Full Article

Mirk/dyrk1B decreases the nuclear accumulation of class II histone deacetylases during skeletal muscle differentiation Dr. Eileen Friedman and her collaborators at Upstate Medical University, State University of New York, Syracuse described in a mouse myoblast model that myogenic transcription factors are regulated by class II HDACs via Mirk kinase signaling. The study used Abgent’s antibody to HDAC9/MITR (Cat.# AP1109a) to monitor MITR cellular distribution. Review Data

| Kinases in Neurodegenerative Diseases |
Featured Article - Protein kinases represent as much as thirty percent of all protein targets under investigation by pharmaceutical companies. Recent successful launches of cancer-targeted drugs with kinase inhibition as the mode of action demonstrate the ability to deliver kinase inhibitors as drugs with the appropriate selectivity, potency, and pharmacokinetic properties. To date, kinase brain research, however, has not yet delivered many candidates for treating neurodegenerative disorders such as Alzheimer and Parkinson diseases. LRRK2 in Parkinson’s Disease and Dementia with Lewy Bodies A project led by Dr. S. Chen at Case Western University in Cleveland, OH described the use of antibodies to label brainstem and cortical Lewy bodies, the pathological hallmarks of Parkinson’s disease (PD) and dementia with Lewy bodies (DLB), respectively. They demonstrated that LRRK2 is a component of Lewy bodies in both PD and DLB, and therefore plays an important role in the Lewy body formation and disease pathogenesis. Review Data

Q: How do I store my antibodies?
A: It is good practice to aliquot your antibodies in small volumes upon receiving. We recommend using multiple aliquots of 20-100 ul for frequent bench work, whereas bulk is stored away in 5-20 ml tubes. Antibodies are robust proteins although multiple freeze-thaw cycles will diminish their properties. They should be frozen at 20°C for long term storage (>1 month), -80°C for crude sera to minimize proteolysis. Small aliquots can be kept at 4°C for frequent use within few days, otherwise at 20°C, especially for crude or partially purified antibodies. Don’t forget that antibodies have general stability properties that can vary from one antibody to another. Stability testing is recommended for unknown antibodies.
