Starting with a customer-supplied antigen, this complete polyclonal antibody production service option produces 2 – 10 mg of antigen affinity-purified antibody using a 12-week protocol. The customer-supplied antigen can be a native or recombinant protein, or a peptide. Purification is an important aspect of all antibody custom service projects. We perform standard Protein A or antigen affinity purification of polyclonal antibodies. Peptide affinity purification is recommended, as it produces high quality monospecific antibodies with enhanced affinity and low background.
12-week protocol includes the following using a customer-supplied antigen:
- Antigen-KLH conjugation
- Immunization of two specific pathogen free (SPF) New Zealand rabbits
- Bleed titration to ensure high immune response
- Protein A purification and antigen affinity-purification
- Options available: additional rabbits; protocol extension; antibody purification using antigen affinity column, Western blot, immunohistochemistry and immunoprecipitation testing
