PKC antibodies specific for isoform nu

In the past two years Abgent provided antibodies to the different isoforms of PKC kinases for evaluation in our laboratory. Products Cat.# AP7025a and AP7025b are directed against the PKC nu isoform , also called PKD3. Products Cat.# AP7730a and AP7730b are directed against the PKD-C2 isoform, also called PKD2.

The PKC nu antibodies could detect GFP-PKD3 overexpressed in Panc-1 cells in western blot and indirect immunofluorescence (see figure). PKD3 was localized in the cytoplasm and nucleus of Panc-1 cells, a distribution observed in our previous work (Rey O. et al. 2003 JBC 278: 23773). As seen in the western blot, AP7025a gives a stronger signal than AP7025b. The PKD2 antibodies recognize both PKD2 and PKD (also called PKC mu) in Western blot (data not shown), therefore no further characterization was performed.

In the last few years the relevance of PKDs in the field of signal transduction is becoming more apparent as revealed by the increase number of publications dedicated to these signaling molecules. I am sure several groups will be delighted to have specific antibodies that recognize the different PKD isoforms.
