  • WB - Anti-Huntingtin Antibody, clone HDC8A4 ABD11684
Total protein extract of normal human cerebral cortex separated as a strip on a 3-12.5% gradient SDS-PAGE gel and Western blotted. The blot was probed with ABD11684
  • IF - Anti-Huntingtin Antibody, clone HDC8A4 ABD11684
Published customer image:Mouse anti Huntingtin antibody, clone HDC8A4 used to demonstrate perinuclear huntingtin expression and it's co-localization with the endoplasmic reticulum by immunofluorescence.Image caption:No increase of Htt co-localization with Rab22A. Representative confocal microscopic immunofluorescent images of Htt and Rab22A in STHdhQ7/Q11 cells and quantitation analysis of co-localization. Treatments with 1 μM RP-cAMP or 1 μM 8-pCPT-2'-O-cAMP for 48 hours leads to no change of co-localization between Htt and Rab22A. Red Rab22A; Blue: DAPI; Green: Htt; Scale bars 20μm.From: Yao Y, Cui X, Al-Ramahi I, Sun X, Li B, Hou J, Difiglia M, Palacino J, Wu ZY, Ma L, Botas J, Lu B. Published customer image:Mouse anti Huntingtin antibody, clone HDC8A4 used to demonstrate perinuclear huntingtin expression and it's co-localization with the endoplasmic reticulum by immunofluorescence.Image caption:Confocal microscopy experiments showing that HTT proteins are enriched in the perinuclear and co-localize with the endoplasmic reticulum marker calreticulin upon treatment of Rp-cAMP or 8-pCpt2'-O-Me-cAMP (8-pCpt-cAMP). upper panels representative images showing the immunofluorescent signals of Htt , ER marker (red, only in the third and fourth columns) and DAPI in STHdh Q7/Q11 cells treated by vehicle, 1 μM Rp-cAMP or 1 μM 8-pCpt-cAMP. Scale bars, 20 μM. The two panels on the right side are magnified images from the left for visualizing the co-localization. Yellow pixels indicate co-localization.From: Yao Y, Cui X, Al-Ramahi I, Sun X, Li B, Hou J, Difiglia M, Palacino J, Wu ZY, Ma L, Botas J, Lu B. Treatment with 1 μM RP-cAMP or 1 μM 8-pCPT-2'-O-Me-cAMP for 48 hours leads to increased co-localization between Htt and Rab39B. Red: Rab39B; Blue: DAPI; Green: Htt; The lower panels are magnified images from the upper for visualizing the co-localization. Yellow pixels indicate co-localization. Scale bars: 20 μm. Right panels: co-localization parameters including Pearson's coefficient and overlap coefficient (mean and s.e.m.). Target/Specificity Mouse anti-Huntingtin antibody, clone HDC8A4 reacts with an epitope corresponding to the HDC region (2703 - 2911 amino acids) of the huntingtin protein.Mouse anti-Huntingtin antibody, clone HDC8A4 detects a 350KDa band on western blots but also detects smaller degradation products of huntingtin. Clone HDC8A4 recognises both denatured and native huntingtin in human brain. The combined use of Mouse anti-Huntingtin antibody, clone HDC8A4 (ABD11684), HDB4E10 (MCA2050) and HDA3E10 (MCA2049) demonstrate that huntingtin is enriched in neuronal cells in the brain.
