Protein Information
Synonyms CD45
Function Protein tyrosine-protein phosphatase required for T-cell activation through the antigen receptor. Acts as a positive regulator of T-cell coactivation upon binding to DPP4. The first PTPase domain has enzymatic activity, while the second one seems to affect the substrate specificity of the first one. Upon T-cell activation, recruits and dephosphorylates SKAP1 and FYN. Dephosphorylates LYN, and thereby modulates LYN activity (By similarity).
Cellular Location Membrane; Single-pass type I membrane protein Membrane raft. Note=Colocalized with DPP4 in membrane rafts

CD45, also known as the leukocyte common antigen (LCA), is a transmembrane protein-tyrosine phosphatase (PTPase) that is expressed in hematopoietic cells except erythrocytes and platelets (1). CD 45 has been implicated in activating, differentiating and the development of the immune system. It regulates T-or B-cell activation during receptor-specific recognition by dephosphorylating tyrosine residues in protein kinase substrates. It is also required for T- and B-cell activation by regulating Lck & Fyn and Lyn & Blk respectively (2). Additionally, the expression of the CD45 5 isoforms can be seen in the various stages of differentiation of normal hematopoietic cells depending on the activation status of the cell (3).
