|Function||Regulates CDK7, the catalytic subunit of the CDK- activating kinase (CAK) enzymatic complex. CAK activates the cyclin-associated kinases CDK1, CDK2, CDK4 and CDK6 by threonine phosphorylation. CAK complexed to the core-TFIIH basal transcription factor activates RNA polymerase II by serine phosphorylation of the repetitive C-terminal domain (CTD) of its large subunit (POLR2A), allowing its escape from the promoter and elongation of the transcripts. Involved in cell cycle control and in RNA transcription by RNA polymerase II. Its expression and activity are constant throughout the cell cycle.|

Cyclin H, a member of the cyclin family, functions as a regulator of CDK kinases (1). It associates primarily with the kinase CDK7 and ring finger protein MAT1 to form the CDK-activating kinase (CAK) complex. CAK binds and activates the cyclin-associated kinases CDC2/CDK1, CDK2, CDK4 and CDK6 by threonine phosphorylation (2). Cyclin H is also a substrate of protein kinase 2, a ubiquitously expressed serine/threonine protein kinase required for cell viability and cell cycle progression (3). In addition, the CAK complex is a component of TFIIH and the RNA polymerase II protein complex (2). It participates in two different transcriptional regulation processes, suggesting an important link between basal transcription control and the cell cycle machinery (1).
