BACKGROUND
Cystatin B is a member of the cysteine protease inhibitor superfamily, functioning as an intracellular thiol protease inhibitor (1). It plays a role in protecting proteinases from lysosomal leakage (1, 2). The second hairpin loop and the C-terminal end of Cystatin B anchor the inhibitor to target proteinases (3). Cystatin B interacts with and inhibits proteases of the cathepsin family (4). A deficiency in cystatin B is observed in progressive myoclonic epilepsy type 1 (1). This deficiency predisposes neurons to oxidative stress that leads to neuronal death and degeneration (1). In addition, Cystatin B has been linked to amyloid fibril formation in vitro and is overexpressed in hepatocellular carcinoma (5, 6).
