|Function||Keeps in an inactive conformation TGFBR1, the TGF-beta type I serine/threonine kinase receptor, preventing TGF-beta receptor activation in absence of ligand. Recruites SMAD7 to ACVR1B which prevents the association of SMAD2 and SMAD3 with the activin receptor complex, thereby blocking the activin signal. May modulate the RYR1 calcium channel activity. PPIases accelerate the folding of proteins. It catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides.|

FKBP12 (Fkbp1a) is a member of the FK-506-binding protein (FKBP) family. This family of proteins possesses peptidyl-prolyl cis-trans isomerase (PPIase) activity and is believed to play an important role in accelerating the rate at which proteins fold into their native conformations (1). FKBP12 was originally identified as a cytosolic receptor for the immunosuppressant drugs, FK506 and rapamycin, and later shown to be involved in cell-cycle regulation and in intracellular calcium homeostasis. Binding of FKBP12 to rapamycin through its FK506/rapamycin-binding/ PPIase domain mediates the immunosuppressive and anti-tumor effects of rapamycin, a drug that has shown to arrest tumor cells in G1 phase of the cell cycle and to reduce tumor angiogenesis (2). FKBP12 is highly conserved from yeast through human and is relatively abundant in most tissues and cell types (3).
