Nuclear factor of activated T-cells, cytoplasmic 2 (NFAT1) belongs to the NFAT family of transcription factors that are expressed in most immune cell types, and play a central role in the transcription of cytokine genes, such as IL-2, IL-4, IL-5, IL-13, IFN-gamma, TNF-alpha, and GM-CSF. The activity NFAT proteins are regulated by the calcium/calmodulin-dependent phosphatase calcineurin, a target for inhibition by CsA and FK506 (1). NFAT1 is present in the cytosol and only translocates to the nucleus upon T cell receptor (TCR) stimulation, where it becomes a member of the NFAT transcription complex. This complex plays a central role in inducing gene transcription during the immune response (2).
