|Function||Plasma membrane-associated small GTPase which cycles between an active GTP-bound and an inactive GDP-bound state. In active state binds to a variety of effector proteins to regulate cellular responses. Involved in epithelial cell polarization processes. Regulates the bipolar attachment of spindle microtubules to kinetochores before chromosome congression in metaphase. Plays a role in the extension and maintenance of the formation of thin, actin-rich surface projections called filopodia. Mediates CDC42-dependent cell migration.|

Rac1 and cell division cycle 42 (cdc42) are guanosine triphosphatases (GTPases). Rac1 is involved in signal transduction pathways that induce the formation of lamellipodia, stimulate cell proliferation and activate the JNK/SAPK protein kinase cascade. Rac1 regulates unique aspects of hematopoietic development and function, and is critical for the hypertrophic response in the heart. Therapies that target myocardial Rac1 are reported to be beneficial in the treatment of cardiac hypertrophy. Cdc42 has been shown in endothelial cell monolayers to prevent disassembly of interendothelial junctions and the increase in endothelial permeability. Cdc42 activity of microvessel endothelial cells is a critical determinant of junctional barrier restrictiveness and may represent a means of therapeutically modulating increased lung vascular permeability and edema formation. The phosphorylation of Rac1 inhibits its GTP-binding activity without any significant change in GTPase activity. Akt phosphorylates serine 71 of Rac1, modulating the Rac1 signal transduction pathway (1-5).
