Cytosolic thymidine kinase 1, TK1, is a well known cell-cycle-regulated enzyme of importance in nucleotide metabolism as well as an activator of antiviral and anticancer drugs (1). TK1 catalyzes the ATP-dependent phosphorylation of thymidine to thymidine 5'-monophosphate, which is subsequently converted to thymidine 5'-triphosphate and utilized for DNA synthesis. In terminally differentiating cells, such as cardiac muscle cells, the cessation of DNA synthesis and withdrawal from the cell cycle are accompanied by the loss of TK1 activity (2). TK1 is a key enzyme of the salvage pathway for pyrimidine deoxyribonucleotide synthesis in a variety of organisms (3). Two forms have been identified in animal cells, one in cytosol and one in mitochondria. Activity of the cytosolic enzyme is high in proliferating cells and peaks during the S-phase of the cell cycle; it is very low in resting cells (4).
