|Other Names||NACHT, LRR and PYD domains-containing protein 10, Nucleotide-binding oligomerization domain protein 8, NLRP10, NALP10, NOD8, PYNOD|
|Target/Specificity||The synthetic peptide sequence used to generate the antibody AP13494b was selected from the C-term region of NLRP10. A 10 to 100 fold molar excess to antibody is recommended. Precise conditions should be optimized for a particular assay.|
|Format||Synthetic peptide was lyophilized with 100% acetonitrile and is supplied as a powder. Reconstitute with 0.1 ml DI water for a final concentration of 1 mg/ml.|
|Storage||Maintain refrigerated at 2-8°C for up to 6 months. For long term storage store at -20°C.|
|Precautions||This product is for research use only. Not for use in diagnostic or therapeutic procedures.|
|Synonyms||NALP10, NOD8, PYNOD|
|Function||Inhibits autoprocessing of CASP1, CASP1-dependent IL1B secretion, PYCARD aggregation and PYCARD-mediated apoptosis but not apoptosis induced by FAS or BID. Displays anti-inflammatory activity. Plays a role in adaptive immunity through control of dendritic cell-mediated transport of antigen to the lymph nodes from peripheral sites. Required for immunity against C.albicans infection. Involved in the innate immune response by contributing to proinflammatory cytokine release in response to invasive bacterial infection.|
|Cellular Location||Cytoplasm. Cell membrane. Note=Cytoplasmic protein which is recruited to the cell membrane by NOD1 following invasive bacterial infection|
|Tissue Location||Highly expressed in basal and suprabasal epidermal cell layers with lower levels in dermal fibroblast cells (at protein level). Widely expressed with highest levels in heart, brain and skeletal muscle. Also expressed in liver, colon, dermis and epidermis. Little expression detected in myeloid cells or peripheral blood mononuclear cells.|

Members of the NALP protein family typically contain aNACHT domain, a NACHT-associated domain (NAD), a C-terminalleucine-rich repeat (LRR) region, and an N-terminal pyrin domain(PYD). The protein encoded by this gene belongs to the NALP proteinfamily despite lacking the LRR region. This protein likely plays aregulatory role in the innate immune system. The protein belongs tothe signal-induced multiprotein complex, the inflammasome, thatactivates the pro-inflammatory caspases, caspase-1 and caspase-5.Other experiments indicate that this gene acts as a multifunctionalnegative regulator of inflammation and apoptosis.
