Protein Information
Name RNF111
Function Acts in the NODAL pathway of mesoderm patterning during embryonic development. Acts downstream AXIN1 as an E3 ubiquitin- protein ligase which promotes the ubiquitination of inhibitory SMADs such as SMAD7, induces their proteasomal degradation and thereby enhances the transcriptional activity of TGF-beta and BMP. Activates Smad3/Smad4-dependent transcription by triggering signal-induced SnoN degradation. Associates with UBE2D2 as an E2 enzyme.
Cellular Location Nucleus. Cytoplasm. Note=Upon TGF-beta treatment, translocates from nucleus to cytosol
Tissue Location Broadly expressed.

The protein encoded by this gene contains a RING fingerdomain, a motif known to be involved in protein-protein andprotein-DNA interactions. The mouse counterpart of this gene(Rnf111/arkadia) has been shown to genetically interact with thetransforming growth factor (TGF) beta-like factor Nodal, and act asa modulator of the nodal signaling cascade, which is essential forthe induction of mesoderm during embryonic development.
