InvolucrinÂ is aÂ proteinÂ component of human skin and in humans is encoded by theÂ IVLÂ gene. It is a highly reactive, soluble,Â transglutaminaseÂ substrateÂ proteinÂ present inÂ keratinocytesÂ ofÂ epidermisand otherÂ stratified squamous epithelia. It first appears in theÂ cellÂ cytosol, but ultimately becomes cross-linked toÂ membrane proteinsÂ by transglutaminase thus helping in the formation of an insoluble envelope beneath theplasma membraneÂ functioning as aÂ glutamylÂ donor during assembly of the cornified envelope. Additionally, Involucrin is synthesised in theÂ stratum spinosumÂ and cross linked in theÂ stratum granulosumÂ by thetransglutaminaseÂ enzyme that makes it highly stable. Thus it provides structural support to the cell, thereby allowing the cell to resist invasion by micro-organisms.
